Before we use Multi-Site on WordPress 3.0, we need to enabled it. How To Enable WordPress MU on WordPress 3.0 is really simple, open you wp-config.php and added this parameter
define('WP_ALLOW_MULTISITE', true);

That’s, after you done with it, you will have new menu on your WordPress Dashboard > Tools, the menu called Network.

Please note, if you want Multi-Site has a sub domain example (http://myblog.wptrikcs.net, http://yourblog.wptricks.net) You need added Wildcard (*) on your DNS configuration. More detailed instruction you can read it on WordPress Codex page here
